of Foxp3+ within the CD25+ after one week of culture We isolated CD4+ T cells from spleen and lymph nodes (LN) of male C57BL/6 mice following the manufacture’s protocol. CD19+ B cells were enriched using
the CD19+ B-cell Enrichment from spleen of male BALB/c mice. The purity of both cell populations was about 97%. Equal amounts of B cells and CD4+ T cells (3×106 cells/ ml) were seeded into each well of a 24 well plate. In the different experimental set-ups the cells were treated with 1μg/ml anti-CD4 mAb (clone YTS 177) and additionally with 1ng/ml rpTGF-β and 0,5nM all-trans Retinoic Acid or 10nM Rapamycin. n = 3–11. Cells were stained on day 7 of primary culture for CD4, CD25 and FoxP3. FoxP3 frequency is shown gated on CD4+CD25+ T cells. Figure S5.
